Not necessarily, it really just depends how the odds match up with what you think they should be (i.e. is there value according to your numbers). You may get the best odds at any time, however professional gamblers will typically try to bet the initial odds/lines ASAP if they are finding value. On the other hand, you might find the best odds right before game time. I guess what I’m trying to say is that there’s not a certain "best time" to get the best odds since they will continue to fluctuate depending on the market, up until game time.
Does the number of people betting on the line, or which way they are betting effect the line? It’s not necessarily the number of people, but rather the amount of money and who’s betting it. Public money (squares) won’t effect the line nearly as much as sharp money/professionals. More public money coming in on one team will slowly move the line throughout the day while sharp action can move the line immediately.

If you could go back in time and put money on one game, fight, race, series, match, etc. which would earn you the most money. For example, betting 100 bucks on the Warriors to beat Dallas in the 2007 NBA Playoffs would have earned you $1,400.
